Madekwe was raised by an ethnically mixed family. Her father is Nigerian and Swiss as well as her mother is English. In 2008, the actress was offered Bambi as the main character on ITV2's show Secret Diary of a Call Girl. Madekwe also starred as the lead character in How to lose friends & betray people that was aired by Sony Pictures Entertainment in 2008. Her theatre credits include Little Sweet Thing by Roy Williams and Flight Path by David Watson.
Madekwe was cast as Ashley Davenport on the ABC drama The Beautiful Life. The two-episode show aired only for two weeks on The CW and then it came to an end. Ashley Davenport was her role in ABC's drama Revenge. The show premiered in May 2013 and it was revealed that Madekwe, who had been a series regular in the two previous seasons, was to leave the series with a guest appearance on the first episode of the new season. In the television series Salem the actress played Tituba from 2014 until 2016. Her long-time friend, Iddo, married Madekwe in June of 2012.
